This phosphoGlycogen Synthase Kinase (GSK) 3 beta Enzyme Immunometric Assay kit is a complete kit for the determination of phosphoGSK3B in cell lysates. Please read the complete kit insert before performing this assay. The kit uses a monoclonal antibody to GSK3B immobilized on amicrotiter plate to bind theGSK33 in the standards or sample. A recombinant GSK3B Standard is provided in the kit. After a short incubation, the excess sample or standard is washed out and a rabbit polyclonal antibody to phosphoGSK3B (Ser 9) is added. This antibody binds to phosphorylated GSK3B (Ser 9) captured on the plate. After a short incubation the excess antibody is washed out and antirabbit IgG conjugated to Horseradish peroxidase is added, which binds to the polyclonal phosphoGSK3B antibody. Excess conjugate is washed out and substrate is added. After a short incubation, the enzyme reaction is stopped and the color generated is read at 450 nm. The measured optical density is directly proportional to the concentration of phosphoGSK3B in either standards or samples.
