Assay Information
An antimouse IL4 coating antibody is adsorbed onto microwells. Mouse IL4 present in the sample or standard binds to antibodies adsorbed to the microwells. A biotinconjugated antimouse IL4 antibody is added and binds to mouse IL4 captured by the first antibody. Following incubation unbound biotin conjugated antimouse IL4 antibody is removed during a wash step. StreptavidinHRP is added and binds to the biotin conjugated antimouse IL4 antibody. Following incubation unbound StreptavidinHRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of mouse IL4 present in the sample or standard. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A standard curve is prepared from 7 mouse IL4 standard dilutions and mouse IL4 sample concentration determined.

Gene
Interleukin 4 (IL4) is a cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells. It is used for dendritic and T cell therapy. Upon activation by IL-4, Th2 cells subsequently produce additional IL-4 in a positive feedback loop. The cell that initially produces IL-4, thus inducing Th0 differentiation, has not been identified, but recent studies suggest that basophils may be the effector cell.
