Assay Information
An antimonkey IL8 coating antibody is adsorbed onto microwells. Monkey IL8 present in the sample or standard binds to antibodies adsorbed to the microwells. A biotinconjugated antimonkey IL8 antibody is added and binds to monkey IL8 captured by the first antibody. Following incubation unbound biotin conjugated antimonkey IL8 antibody is removed during a wash step. StreptavidinHRP is added and binds to the biotin conjugated antimonkey IL8 antibody. Following incubation unbound StreptavidinHRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of monkey IL8 present in the sample or standard. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A standard curve is prepared from 7 monkey IL8 standard dilutions and monkey IL8 sample concentration determined.

Interleukin 8 (IL8 or chemokine (C-X-C motif) ligand 8, CXCL8) is a chemokine produced by macrophages and other cell types such as epithelial cells, airway smooth muscle cells and endothelial cells. Endothelial cells store IL-8 in their storage vesicles, the Weibel-Palade bodies. In humans, the interleukin-8 proteins encoded by the IL8 gene. IL-8 is initially produced as a precursor peptide of 99 amino acids long which then undergoes cleavage to create several active IL-8 isoforms. In culture, a 72 amino acid peptide is the major form secreted by macrophages. Recombinant, rec. E. coli interleukin-8 for cell culture or antibody production.
Description
