Assay Information
An antihuman IL8/NAP1 coating antibody is adsorbed onto microwells. Human IL8/NAP1 present in the sample or  standard binds to antibodies adsorbed to the microwells. A biotinconjugated antihuman IL8/NAP1 antibody is added and binds to human IL8/NAP1 captured by the first antibody. Following incubation unbound biotin conjugated  antihuman IL8/NAP1 antibody is removed during a wash step. StreptavidinHRP is added and binds to the biotin conjugated antihuman IL8/NAP1 antibody. Following incubation unbound StreptavidinHRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to  the amount of human IL8/NAP1 present in the sample or standard. The reaction is terminated by addition of acid and  absorbance is measured at 450 nm. A standard curve is prepared from 7 human IL8/NAP1 standard dilutions and human IL8/NAP1 sample concentration determined.

Interleukin 8 (IL8 or chemokine (C-X-C motif) ligand 8, CXCL8) is a chemokine produced by macrophages and other cell types such as epithelial cells, airway smooth muscle cells and endothelial cells. Endothelial cells store IL-8 in their storage vesicles, the Weibel-Palade bodies. In humans, the interleukin-8 proteins encoded by the IL8 gene. IL-8 is initially produced as a precursor peptide of 99 amino acids long which then undergoes cleavage to create several active IL-8 isoforms. In culture, a 72 amino acid peptide is the major form secreted by macrophages.
