Assay Information
An antimouse sCD40L coating antibody is adsorbed onto microwells. Mouse sCD40L present in the sample or standard binds to antibodies adsorbed to the microwells. A biotinconjugated antimouse sCD40L antibody is added and binds to mouse sCD40L captured by the first antibody. Following incubation unbound biotin conjugated antimouse sCD40L antibody is removed during a wash step. StreptavidinHRP is added and binds to the biotin conjugated antimouse sCD40L antibody. Following incubation unbound StreptavidinHRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of mouse sCD40L present in the sample or standard. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A standard curve is prepared from 7 mouse sCD40L standard dilutions and mouse sCD40L sample concentration determined.
