An antimonkey IL2 coating antibody is adsorbed onto microwells. Monkey IL2 present in the sample or standard binds to antibodies adsorbed to the microwells. A biotinconjugated antimonkey IL2 antibody is added and binds to monkey IL2 captured by the first antibody. Following incubation unbound biotin conjugated antimonkey IL2 antibody is removed during a wash step. StreptavidinHRP is added and binds to the biotin conjugated antimonkey IL2 antibody. Following incubation unbound StreptavidinHRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of monkey IL2 present in the sample or standard. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A standard curve is prepared from 7 monkey IL2 standard dilutions and monkey IL2 sample concentration determined.

Interleukin-2 (IL-2) is an interleukin, a type of cytokine signaling molecule in the immune system. It is a protein that regulates the activities of white blood cells (leukocytes, often lymphocytes) that are responsible for immunity. IL-2 is part of the body's natural response to microbial infection, and in discriminating between foreign ("non-self") and "self".
