Assay Information
An antihuman sVCAM1 coating antibody is adsorbed onto microwells. Human sVCAM1 present in the sample or standard binds to antibodies adsorbed to the microwells. A conjugate mixture (biotin conjugated antihuman sVCAM1 antibody and StreptavidinHRP) is added. Biotinconjugated antihuman sVCAM1 antibody binds to human sVCAM1 captured by the first antibody. StreptavidinHRP binds to the biotin conjugated antihuman sVCAM1 antibody. Following incubation unbound StreptavidinHRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of human sVCAM1 present in the sample or standard. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A standard curve is prepared from 6 human sVCAM1 standard dilutions and human sVCAM1 sample concentration determined.
