Assay Information
An antihuman IL4 coating antibody is adsorbed onto microwells. Human IL4 present in the sample or standard  binds to antibodies adsorbed to the microwells. A biotinconjugated antihuman IL4 antibody is added and binds to human IL4 captured by the first antibody. Following incubation unbound biotin conjugated antihuman IL4 antibody is  removed during a wash step. StreptavidinHRP is added and binds to the biotin conjugated antihuman IL4 antibody. Following incubation unbound StreptavidinHRP is removed during a wash step, and substrate solution reactive with HRP is added to the wells. A coloured product is formed in proportion to the amount of human IL4 present in the  sample or standard. The reaction is terminated by addition of acid and absorbance is measured at 450 nm. A  standard curve is prepared from 7 human IL4 standard dilutions and human IL4 sample concentration determined.

Interleukin 4 (IL4) is a cytokine that induces differentiation of naive helper T cells (Th0 cells) to Th2 cells. It is used for dendritic and T cell therapy. Upon activation by IL-4, Th2 cells subsequently produce additional IL-4 in a positive feedback loop. The cell that initially produces IL-4, thus inducing Th0 differentiation, has not been identified, but recent studies suggest that basophils may be the effector cell. It is closely related and has functions similar to Interleukin 13.
